SRX1725576: Snodgrassella alvi wkB2, tn-SEQ experimental library replicate 3

Design: tn-SEQ library of Snodgrassella alvi strain wkB2 (bearing a gentR cassette from the Mariner transposon from pBT20) recovered from dissected ileums of 50 Apis mellifera individuals after 5 days post inoculation. gDNA was isolated via a CTAB/bead beating method, fragmented to ~ 300 bp used as a template for C-tailing via TdT. Transposon junctions in C-tailed template were amplified with a transposon specific primer paired with a poly G biotinylated primer. Amplicons bearing insertion junctions were magnetically pulled down, barcoded and adapted for illumina sequencing via PCR. Amplicon was sent for sequencing at the University of Texas at Austin GSAF.
